Brief Profile
I was part of the multiple research programs of the department focusing on early detection and prevention of oral cancer. As a part of the DBT- Wellcome Trust India Alliance-Research Training Fellowship Program, I was working on “Molecular cytology of oral potentially malignant and malignant lesions” wherein the primary goal is to improve on oral cytology as a detection method by including molecular markers.
In addition, I was part of the 5 year NIH grant (UH2EB022623) on “Low cost oral mobile cancer screening for low resource setting” initiated in 2016 in collaboration with University of Arizona” as a Co-Investigator. I was involved in hypothesis generation and grant writing and work has contributed towards the drafting of the proposal “Phase IIb/III study to determine efficacy of Curcumin and Metformin to reduce the incidence of second primary tumors of aero-digestive tract in patients with history of head and neck squamous cell carcinoma” a National Cancer Grid funded clinical trial that is currently ongoing. This is in addition to the successful sanction of the RTF fellowship by the Wellcome Trust DBT India Alliance.
During my previous association with the research program in head and neck oncology, I was involved in evaluating OCT-based (NIH-DBT funded; A Low-Cost Simple Oral Cancer Screening Device for Low-Resource Setting) and fluorescent lectin-based imaging for early detection and/or surgical margin delineation. Additionally, I was instrumental in the efforts initiated in the lab for mobile health, both for mobile phone based imaging (Oncogrid- A mobile health (m-Health) approach to prevention and early diagnosis of oral cancer in Rural south India; collaborated with Boston University) and for telecytology.

Tele- cytology and Artificial Intelligence for cancer detection. mHealth extended to telecytology (Cellscope) that transmit cytology images to connect cytolopathologist with frontline health workers. With collaboration with IISc we developed automated image analysis pipelines and trained ANN based image classification.

Molecular Imaging: We have demonstrated that carcinogenesis results in altered glycan expression and nuclear damage on oral cancer cells. Paired normal and tumor tissue from oral squamous cell carcinoma, oral pre-cancer and benign lesions diagnosed were investigated for sialic acid expression via the legume protein wheat germ agglutinin (WGA)/PariFL. Fluorophore conjugated WGA/ParpiFL was topically applied to the tissue samples and imaged using a custom wide-field fluorescence imaging system.

Chemoprevention: We have demonstrated that cancer initiating cells (CIC) and their niche plays a role during progression of oral dysplastic lesions to invasive carcinoma. Subsequently, we have then demonstrated in a cohort of patient with oral potentially malignant lesions (OPML) that both CIC as well as its niche marker (SDF-1, CXCR-4, CD31) expressions increases with higher grades of dysplasia of OPML. In addition, exposure of oral dysplastic cells to tobacco extract increased the expression of oral cancer CIC (CD44, CD133, ALHD1A1) as well as proliferation (Ki67) and differentiation (CK-14) markers. Treatment of these cells with oral cancer chemo preventive agents (Retinoids and Curcumin) led to abolition of the expression of proliferation and differentiation markers, however only partially suppressed CIC markers.
